Webinar David Henry, Study Monitor at AstraZeneca UKThis presentation will provide an overview of the use of cells in curative treatments (highlighting the use of Heart Ventricular Progenitor Cells in heart failure) and considerations for monitoring outsourced studies supporting first time in human (FTIH) trials. This nonclinical model presents significant study design and practical challenges that need addressing to ensure successful xenogeneic engraftment following intra cardiac dosing in minipigs. In addition, the implementation of an appropriate immunosuppression regimen is essential for successful engraftment of cells, for which it is important to confirm procurement of co-medications, navigating the dosing of multiple drugs and monitoring/maintenance of exposure of the comedication. Therefore, an overview of study designs, immunosuppression, the challenges of intra-cardiac dosing cells and supporting animals with appropriate welfare practices will be discussed.

In this advanced learner’s course, you will learn about Göttingen Minipigs' biology and behaviour. We will go into detail with animal welfare aspects tailored to fit the specific needs of Göttingen Minipigs, incl. valuable acclimatisation, socialization, and thorough behavioral management programs addressing various enrichment elements etc. We will also discuss the importance of working with a Culture of Care, and suggestions on how to promote it. We will take a look at clinical issues that you might encounter in Göttingen Minipigs, as well as their suggested treatment protocols. You will learn about how to maintain healthy minipigs, incl. how to set up effective biosecurity measures and perform adequate health monitoring. To complete the course, a workshop will put the acquired learnings into perspective.

Carina has worked with taking care of Göttingen Minipigs for more than 17 years. Since 2020, she has been responsible for the constant focus on further improving the life and welfare of all animals at Ellegaard Göttingen Minipigs.

Pig vascular access can be a challenge due to the general lack of superficial veins. Göttingen Minipigs are no exception and careful consideration has to be given already during the design process of the study. In this course, we will present the possibilities and tools that can be applied to make the study a success.
After an overall introduction to Göttingen Minipigs covering behavior, biology, and basic handling we will look closer at the vascular anatomy. That will lead to the practical and technical aspects, from simple blood sampling by venipuncture to implantation of catheters and ports. To round off, we discuss the benefits of different systems and what effect they may have on animal welfare.
At the end of the course, the participant should be able to make an informed decision about what option will work best for a given study. The technical insights of this seminar will provide the basics of catheter implantation and the experienced participant will be able to perform the procedure afterward.

About the lecturer: Adrian has worked with Göttingen Minipigs since 2005, and now works as a consultant for AZE Consulting, where his primary focus is on surgical procedures. In addition to his practical background, Adrian has conducted animal training courses and workshops for many years. Throughout his career, Adrian has showcased his surgical proficiency by successfully performing numerous procedures on Göttingen Minipigs including enhancement and innovation of dosing techniques and surgical methodologies.

How to Train Your Göttingen Minipigs
Ellegaard Göttingen Minipigs A/S | Dalmose, Denmark Dorte Bratbo Sørensen, DVM, PhDGöttingen Minipigs are incredibly intelligent creatures and require well-thought-of stimuli to thrive. They are highly trainable and love a good challenge. Every time you train your Göttingen Minipigs, you are also improving your understanding and communication with the animal. Correct handling and training, therefore, not only minimizes stress and improves minipig welfare but also makes the work with minipigs more rewarding and the data they generate more reliable.
In this course, you will learn the theory of different training methods like classical and operand conditioning as well as other training methods. We discuss how this can benefit your studies and which limitations there might be. You will also get a chance to boost your skills as the theoretical lectures will be interrupted with practical training with clickers. With the support of videos, you will be shown examples of what can be achieved with good and planned training.
This is a practical course with limited seatings and designed for novices or experienced animal trainers without any pre-existing knowledge or experience with minipigs. After the theoretical program, each participant will have the possibility to train with their own minipig with the lecturer on the side.

About the lecturer: Dorte is an Associate Professor at the Department of Veterinary and Animal Sciences at the University of Copenhagen and has a PhD in ethology and animal welfare. She specialises in laboratory animal welfare and ethology, animal ethics, 3Rs, and Culture of Care, and her main research areas revolve around the use of positive reinforcement to train laboratory animals to voluntarily participate in procedures and enhancement of refinement through animal-centric management, husbandry, and procedures.

The tasks involved in handling these live animals often require repetitive movements, heavy lifting, and maintaining awkward positions, which can lead to pain and work-related musculoskeletal disorders.
However, this course is specifically designed to address these challenges and teach you how to perform your work while prioritizing your physical well-being.
In the world of ergonomics, many things have come to light over the last 10 years. For example, we now know, after extensive research in the field of ergonomics, that the most important thing is that you experience your daily work as overall comfortable regardless of the ergonomics. Also, as little as two minutes of daily exercise with an exercise band keeps your body in good shape for work.
Led by our experienced educator, this course will highlight the importance of overall physical well-being in the context of working with animals. Throughout the course, you will receive practical tips and tricks on how to practice healthier work positions specifically tailored to the unique demands of working with animals. You will learn proper body mechanics, lifting techniques, and ergonomic strategies to reduce the risk of injuries and discomfort. In order to provide a hands-on learning experience, this course also incorporates interactive lectures and practical exercises. Every participant will also be given an exercise band and learn specific exercises which you can revisit on video after the course.
Whether you are involved in the care of large or small laboratory animals, this course is tailored to address the specific challenges you may encounter in your daily work. About the lecturer: For more than 10 years, Søren has worked to help people obtain a better physical work environment and thus better well-being. Get an introduction on how to create an enriched habitat that supports the biological needs of Göttingen Minipigs. Engage in hands-on learning as you approach, lift, and carry the minipigs under supervision. Try out various restraint techniques and different dosing techniques including oral dosing. Discover the practicality of slings and perform blood sampling within them. Depending on the participants’ level of experience, there will also be room for more specific individual training like endotracheal intubation and catheterization methods for vascular access. Humane euthanasia will also be taught on the course.
This is a practical course with limited seatings, so each participant gets plenty of possibilities to work on their own technique and train in a variety of procedures. By course end, you'll possess the ability to work confidently and empathetically with Göttingen Minipigs.

Humanized Göttingen Minipigs for the toxicological testing of therapeutic antibodies
The safety and efficacy of most novel biologicals is evaluated in animal models. However, most human therapeutic antibodies trigger xenogeneic responses in wild-type animals and thus rapid clearance of the drugs, which makes in vivo testing of human antibodies challenging. Here we report the generation of Göttingen Minipigs carrying a mini-repertoire of human genes for the immunoglobulin heavy chains γ1 and γ4 and the immunoglobulin light chain κ. In line with observations in human patients, the genetically modified minipigs tolerated the clinically non-immunogenic IgG1κ-isotype monoclonal antibodies daratumumab and bevacizumab, and elicited antibodies against the checkpoint inhibitor atezolizumab and the engineered interleukin cergutuzumab amunaleukin. The humanized minipigs can facilitate the safety and efficacy testing of therapeutic antibodies.

Alzheimer’s disease (AD) affects an estimated 40 millions patients worldwide and is still without any cure despite decades of research and billions of dollars spent trying to develop pharmacological interventions. The reasons for this failure are heavily debated in the field, but there is consensus that part of the explanation is the translational gap between promising studies conducted in cells and mouse models and clinical trials aiming at curing or alleviating this complex brain disorder in humans. Göttingen Minipigs - a new model for gut microbiome studies
In 2021, a business/university consortium received funding from the Innovation Fund Denmark to develop translational animal models for inflammatory bowel disease in man. The focus will be on induction of intestinal inflammation and characterising the dynamics of the gut microbiome of various pre- and probiotic intervention. The project combines two omnivore animal models (insects: Fast and inexpensive screening; Minipigs: Highly predictive) for preclinical research on pipeline formulations in biotech and pharma intended for microbiome stimulation and treatment of chronic intestinal disorder in humans. In this presentation preliminary observation will be presented by Professor Christian Kapel, University of Copenhagen, Denmark. Please find more information about the project on www.minigut.dk

Minipig inhalation is an emerging field in toxicology due to their homology to humans. In this webinar, Alice McNamara MRCVS from Labcorp will share how the Labcorp team has successfully completed several regulatory toxicology studies using this model, and have implemented multiple welfare refinements including: • Training. Design of a bespoke positive reinforcement training protocol to acclimatise animals to staff, training them to walk in a harness to the dosing suite and then tolerate wearing a mask for up to 120 minutes/session. • Physical environment. From 3D printing masks in a variety of sizes to give a comfortable fit and minimise dead space, to optimising the dosing suite for pig comfort with tactile, auditory and olfactory enrichment, the environment of the dosing suite allowed animals to calmly settle and even fall asleep during dosing procedures. • Safety monitoring. The safety of the animals is paramount in any study, so capnography and visual scoring to monitor the animals continuously was employed.

Göttingen Minipigs as a Model to Evaluate Longevity, Functionality and Memory of Immune Response Induced by Pertussis Vaccines
Evaluation of the short-term and long-term immunological responses in a preclinical model that simulates the targeted age population with a relevant vaccination schedule is essential for human vaccine development. A neonatal and infant Göttingen Minipig model using whole cell and acellular pertussis vaccines was developed to assess antigen-specific humoral and cellular responses in a longitudinal study. The main immune parameters tested included total IgG, functional antibodies, Th cell subsets and memory T and B cells. Single cell RNA sequencing was used to explore the long-term maintenance of circulating immune memory cells and their functionality. Overall, the minipig model reproduced the immune responses induced in humans by whole cell versus acellular pertussis vaccine priming, followed by acellular pertussis vaccine boosting. Associate Scientist Céline Vaure, from Sanofi Pasteur in France, will present results suggesting that Göttingen Minipigs are an attractive preclinical model to predict the long-term immunogenicity of human vaccines against Bordetella pertussis and that it could be of further interest for vaccines against other pathogens.

There has been a slow uptake to use minipigs as an alternative second species for inhalation studies, which is in part due to technical challenges, but also the historical classes of test articles being used such as beta-agonists, corticosteroids, anti-muscarinic for asthma and COPD, where the dog has been the species of choice based on available background data. However, with ongoing focus and debate on second species selection actively requesting the industry to equally consider all species and in many ways especially minipigs, there is now greater support for the minipig to be used more frequently. Global Lead of Inhalation Sciences and Engineering, Simon Moore from Covance will discuss this opportunity, which is further supported by evidence, where the inhalation delivery route is now being used for respiratory diseases such as cystic fibrosis and idiopathic pulmonary fibrosis with novel compound classes or re-purposing from other routes of administration.

Acute hypersensitivity reactions to intravenously administered nano-biopharmaceuticals, also known as Infusion Reactions (IRs), are complex, innate immune reactions arising from a variable combination of complement (C) activation and triggering of pattern recognition receptors on allergy mediating cells. Pigs, due to the presence of intravascular macrophages in the microcirculation of their lungs, called PIM cells, provide a sensitive model for studying the most dangerous, occasionally life-threatening cardiovascular response leading to anaphylactoid shock and death. Janos Szebeni, CEO at SeroScience Ltd. in Hungary, presents highlights of the clinical and pharmaco-industrial relevance of this problem and discusses the benefits of pigs compared to other animals in consistently reproducing human symptoms. Beyond detailing the conditions, mechanism, and prevention of IRs clarified in domestic pigs, the presentation will give an account of experiments in Göttingen Minipigs that suggest their suitability as a model for drug-induced IRs with all advantages for regulatory standardization. Other focal points will include immunogenicity of nano-biopharmaceuticals and anti-PEG antibodies.

Safety pharmacology, toxicology and pharmacokinetic (PK) data are critical in drug development to provide safety and exposure information for the clinical studies. For optimal species selection in the nonclinical studies, metabolite profiling and plasma protein binding are key PK data to be considered. Metabolites can be investigated in several in vitro matrices, including hepatocytes, subcellular fractions of liver (microsomes, S9), recombinant enzymes, as well as in vivo. As all of these in vitro matrices, except recombinant enzymes, are commercially available for Göttingen Minipigs, it is surprising that this species is still not routinely included in the in vitro drug metabolism testing battery of most pharmaceutical companies. In this presentation, Steven Van Cruchten, Professor in Comparative Perinatal Development at the University of Antwerp, will focus on drug metabolism in adult and juvenile minipigs together with their relation to other non-rodent species and man.

Induction of prediabetes and diabetes in adult obese Göttingen Minipigs
To follow the natural course of obese type 2 diabetes in an adult Göttingen Minipig model, the pig needs to become obese first and subsequently develop diabetes. Once insulin-resistant obesity is established in adult Göttingen Minipigs by ad libitum feeding of a high energy Western diet, diabetes should evolve after a certain period of time. The capacity of pancreatic beta-cells to produce sufficient insulin to compensate for insulin resistance becomes limited and, as a result, blood glucose concentrations increase. However, pigs do not easily develop (pre)diabetes spontaneously, at least not in the time frame of 1 or 2 years after the induction of severe obesity. To accelerate the development of diabetes in obese Göttingen Minipigs, streptozotocin can be used to chemically reduce the pancreatic beta-cell mass in order to elevate blood glucose concentrations. In this webinar, Sietse Jan Koopmans from Livestock Research at Wageningen University & Research in The Netherlands, will discuss various strategies to induce prediabetes and diabetes in adult obese Göttingen Minipigs by repeated intravenous administration of streptozotocin.

The use of Minipigs in juvenile toxicity testing
Göttingen Minipigs have become popular as a non-rodent species in routine repeat dose safety testing during the last few years and is also a promising model for juvenile toxicity studies. For pediatric indications requiring juvenile toxicity testing, the rat is still the preferred species because it is the most convenient and widely studied species with extensive historical control data and well-established testing methods. However, for some drugs it might not be an appropriate model or regulatory agencies may request both rodent and non-rodent juvenile studies, for example in case of pediatric-only indications. The minipig represents good similarity to man in terms of many organ systems, including their post-natal development. The most common non-rodent species for juvenile studies is still the dog, but the minipig has many practical advantages over the dog and monkey. Due to the relevant recent use of minipigs in this field, only a limited number of studies have been conducted within each age category (with commencement of dosing at different ages) and therefore background data are limited. In this webinar, Bianca Feyen, Toxicologist at Janssen in Belgium, will discuss the advantages of using minipigs in juvenile toxicity testing and provide an overview of available background data.

Julia Lodder-Gadaczek: For the development of a transdermal therapeutic system (TTS) in vitro permeation experiments are used as models for the penetration of a drug through the skin to the vesicular system. These experiments are necessary to generate information about the properties of a new formulation in terms of drug penetration, kinetic profile of permeation, and needed concentration levels of the drug. The barrier in this model is normally built by skin and for the early development stage of a new TTS the skin of Göttingen Minipigs is a useful model. In this webinar, the principle of the experiment, the preparation of the skin and the advantages and potential risks of using Göttingen Minipigs skin as a model for in vitro permeation trials will be discussed. Anette Sams Nielsen: Minipigs are superior animal models for human health and diseases with cardiovascular anatomy and physiology similar to humans. Rodent models of neurovascular and endothelial regulation of vascular tone have been thoroughly studied in vivo and ex vivo and do not always offer good translation to sparse human vascular biopsies available from surgery and biopsies. The size of Göttingen Minipigs allows us to dissect sufficient artery segments for thorough characterization, pairing and comparison using a very limited number of animals (3Rs). The aim of the current study was to characterize regulation of vascular tone in Göttingen Minipigs by selected agonists for sympathetic, parasympathetic, sensory and endothelial pathways and to compare vasomotor responses of coronary, cerebral and peripheral arteries using myographs. We will share our data and discuss the scientific and practical issues of studying minipig and rodent vasomotor actions.

Pigs are of interest in human nutritional research because of similarities in the genome, anatomy, digestive physiology, and body size to humans. Pigs are therefore increasingly being used to understand the exact mechanisms behind the role in progression or prevention of disease. Like humans, pigs are omnivores, and in an experimental setting, they can be fed human-like energy-dense diets high in refined carbohydrates and fat and low in dietary fiber. The domestic pig is relevant for studying digestion, bioconversion, and absorption of diet-derived metabolites, but for the metabolic response to dietary intervention in relation to obesity-related diseases, miniature pigs serve better models. Using Göttingen minipigs at Ellegaard to test a new antiviral (anti-SARS-CoV-2) inhalation drug
A new anti-viral, including anti-SARS-CoV-2, inhalation drug has been tested for safety at Ellegaard Göttingen Minipigs A/S in a collaboration between Ellegaard Göttingen Minipigs, University of Copenhagen, and the Norwegian company (owning the patent) SoftOx. First the drug was tested in an intubated minipig model to maximize lung exposure; the animals were thus anaesthetized and received the nebulised drug once daily for five days. Subsequently a masked inhalation model in slightly sedated minipigs was developed to also test upper airway exposure. Göttingen Minipigs are relevant models for inhalation therapy as they in many ways resemble human respiration physiology and anatomy. Mild dose-dependent changes were seen on histopathology in the highest dosing groups. During the experiments all animals were clinical well and tolerated the experiment fine. Experimental design and experiences are presented by Professor Thomas Bjarnsholt, University of Copenhagen, also consultant for SoftOx, and DVM and postdoc Elin Jørgensen, University of Copenhagen (SoftOx grant).

Xenotransplantation is under development in order to alleviate the shortage of human transplants. For numerous reasons, pigs are the best donor animals. However, the use of pig cells or organs may be associated with the transmission of zoonotic viruses. In this webinar, Dr. Joachim Denner, from Institute of Virology Free University Berlin in Germany, explains the extensive analysis of Göttingen Minipigs, which will be used for the treatment of human diabetes patients in Germany in near future. Using sensitive PCR-based and immunological methods, in some animals hepatitis E virus (HEV), porcine cytomegalovirus (PCMV), and porcine circovirus 2 (PCV2) were found. In addition, porcine endogenous retroviruses (PERV-A, -B, and -C) were found integrated in the genome of all Göttingen Minipigs. However, when islet cells from Göttingen Minipigs were transplanted into cynomolgus monkeys, no transmission of PERV and other porcine viruses was observed. Whereas HEV, PCMV, and PCV2 can be successfully eliminated by antiviral drugs, early weaning, or a vaccine, first experiments to inactivate PERVs using CRISPR/Cas have been reported.

The year of 2019 marks the first 50 Years with Göttingen Minipigs since the Georg-August-University in Göttingen, Germany, managed to establish the first barrier-bred population of Göttingen Minipigs at the University’s experimental farm in Relliehausen, Germany.
Ellegaard Göttingen Minipigs is the global breeder and developer of Göttingen Minipigs, and on behalf of our whole team, I am pleased to invite users of and friends with an interest in Göttingen Minipigs to this Scientific Symposium in celebration of the first 50 years with Göttingen Minipigs.

They are highly trainable and love a good challenge. Every time you train your Göttingen Minipigs, you are also improving your understanding and communication with the animal. Correct handling and training, therefore, not only minimizes stress and improves minipig welfare but also makes the work with minipigs more rewarding and the data they generate more reliable.
In this course, you will learn the theory of different training methods like classical and operand conditioning as well as other training methods. We discuss how this can benefit your studies and which limitations there might be. You will also get a chance to boost your skills as the theoretical lectures will be interrupted with practical training with clickers. With the support of videos, you will be shown examples of what can be achieved with good and planned training.
This is a practical course with limited seatings and designed for novices or experienced animal trainers without any pre-existing knowledge or experience with minipigs. After the theoretical program, each participant will have the possibility to train with their own minipig with the lecturer on the side.

Lecturer: Dorte Bratbo Sørensen, DVM, PhD | University of Copenhagen
About the lecturer: Dorte is an Associate Professor at the Department of Veterinary and Animal Sciences at the University of Copenhagen and has a PhD in ethology and animal welfare. She specialises in laboratory animal welfare and ethology, animal ethics, 3Rs, and Culture of Care, and her main research areas revolve around the use of positive reinforcement to train laboratory animals to voluntarily participate in procedures and enhancement of refinement through animal-centric management, husbandry, and procedures.

How to get to Dalmose: Ellegaard Göttingen Minipigs is located in the beautiful countryside, near Slagelse city with excellent international train connections.
